The Anchor Point South Coast rider was injured during stage three of the National Road Series event in Port MacDonnell on Thursday.
Lane completed 18 kilometres of the 137.6km road race circuit before he fell.
His older brother Matt, who is also completing the five-day tour, finished 44th.
Cyclists battled heavy rain and strong winds during stage three.
Teenage competitor Jesse Coyle will take the general classification lead into stage four – a 92km Heywood to Casterton race – despite suffering the signs of hypothermia.
The New South Wales-based Coyle, riding for the Mobius Future racing team, finished second to stage winner, Drapac Pat’s Veg competitor Liam White.
Portland’s Dylan Lindsey was 75th.
