Police said the pothole was on the Henty Highway about 50 metres north of the turn off with Macleod Street.
"It's a significant road hazard, especially in the dark because the area is not well lit. You want to change lanes and not even drive through it because it's so deep," an officer said.
It's understood the road surface was recent resurfaced but has broken up in the past week during wet weather.
Traffic from Warrnambool and east of Portland will come across the pothole when they turn from the Princes Highway onto Henty Highway and drive into Portland on the 80km/h double lane south-bound stretch.
Drivers have contacted the Portland police station to warn of the danger and it's expected the Glenelg Shire Council and VicRoads will be urged to take immediate action on Monday morning.
