The two-day tour will take in Port Campbell, Peterborough and Wangoom and includes three stages, a 12.7km time trial, a 51km road race on Saturday and a criterium on Sunday morning.
Lobigs, 27, claimed honours in the season’s opening event in Gippsland in February and is set to begin favourite for the Tour of South West.
Her team also began the season with a bang, taking team rankings lead after the Gippsland event.
Sydney’s Ashlee Ankudinoff, 27, is also looming as a contender following a silver medal at the Track World Championships.
She is set to take the reins as team leader in the youth-packed New South Wales Institute of Sport’s Sydney Uni Women's Cycling Team.
For the Wangoom time trial, riders are expected to cross the finish line from 8.30am onwards. The road race, held in Wangoom, is expected to wrap up at about 1pm. The Peterborough leg, on Sunday, will finish at 12.15pm.
