Ennis will lead out three key chances in the eight heats of the group two 450m feature with Matt Machine (heat four, box two), Zinzan Brooke (heat five, box one) and Judicial Man (heat six, box four).
She said Zinzan Brooke looked her standout chance after drawing perfectly.
But, the fifth cup heat is likely to be the most hotly contested of the eight heats – the depth of the field is worthy of a final.
The field includes track record holder Unlawful Entry (box four, 20 wins, $280,000 in prizemoney), Invictus Rapid (box five, 22 wins, $130,000) and Jesaulenko (box seven, 29 wins, $315,000).
Zinzan Brooke has won 15 of just 21 starts and the chaser possesses explosive early speed. "Our dogs have all drawn pretty well and are capable of winning their heats," she said.
"Zinzan Brooke has the red and is in really good form after winning at Shepparton on Friday night.
"He looks like leading and can clock some outstanding early splits. His 24.90 run at Shepparton shows what he can do. You don't see that sort of time run at Shepparton very often."
Ennis said Unlawful Entry would be hard to hold off – if he got a clear run early.
She said Judicial Man was also likely to lead with recent group one Golden Easter Egg winner Striker Light the biggest danger to run him down.
The eight heats of the cup and four semi-finals of the Warrnambool Classic kick off at 7pm.
