Buildings 11, 13, 55, 56 and 57 were mentioned in an email to the university, which threatened violence.
Chief Operating Officer Dionne Higgins said the buildings were temporarily closed as a precaution.
It is understood security sweeps were carried out.
Students sitting exams in the affected buildings will have their exams rescheduled.
The university alerted staff to the threat about 2.20pm on Tuesday, advising them to make alterative working arrangements for the day.
Ms Higgins said the evacuations were necessary.
"RMIT regrets any inconvenience that this closure may cause but we take health and safety very seriously," she said.
Victoria Police confirmed it was conducting safety checks at an educational facility in the CBD.
"As a precaution several buildings were evacuated," a spokeswoman said in a statement.
"As it is currently under investigation, it would not be appropriate to comment further."
It comes after a hoax bomb threat swept state schools across the country and New Zealand on Friday last week, forcing schools to be evacuated and checked by local police.
