The new deal will see the quick-thinking defender remain at the Bombers until at least the end of the 2018 season.
Gleeson said the players had built a strong bond under the guidance of John Worsfold.
“We are demanding elite standards of each other and building a really strong team culture,” he said.
“We know we have plenty of hard work ahead of us if we want to compete with the elite sides in the competition, but we also have so much to look forward to in 2017 and this is a team I want to be part of.
“I’ve really enjoyed my first season under Woosha and I’m excited to be continuing my career at Essendon.”
The 22-year old played 20 games in his fourth season with the club after notching 22 in 2015.
