Police are redirecting traffic along Wollaston Road, Bridge Road and Tower Hill Road.
An ambulance is on the scene and at least one driver is injured but it is unknown at this stage how serious the accident is.
It is not known how many vehicles were involved or how long the road will remain blocked.
The accident occurred in a 100km/h zone.
UPDATE:
Caramut Road has been reopened following a two-car collision that occurred between a small green Mitsubishi vehicle and a four-wheel-drive.
A female driver of one of the vehicles has been taken to hospital.
Police said a tanker had been pulling out onto the road and traffic had been halted.
They said the first vehicle stopped to allow the tanker to enter the road and the second vehicle had collided with the first.
Senior sergeant Diane Grant from the Warrnambool Police warned drivers to pay attention and not to get distracted.
“On country roads you’ve got to be aware,” she said.
The road was closed for approximately 45 minutes.
