Both sides made four changes at the selection table on Thursday night.
The Eagles lost key midfielder Jye Bidmade (unavailable), strong-marking forward Josh Corbett (unavailable) and teenage duo Zac Urquhart and Dion Johnstone, who have been called up for TAC Cup duties with North Ballarat Rebels and Calder Cannons respectively.
Goal sneak Tyrone Bean (hamstring), utlilty Liam Howarth (unavailable) and defensive duo Adam Farley (unavailable) and Mason Crosier (hamstring) will miss for the Seagulls.
Eagles co-coach Brendan Murfett said his side's omissions were "massive outs".
"Josh has been an absolute superstar for us, and Jye as well," he said.
"We have a bit of depth in our under 18s and reserves."
Warrnambool veteran Travis Graham (back) headlines the Blues' changes for their Sunday clash against South Warrnambool.
Kurt Lenehan (knee), Nick Chirnside (hamstring) and Luke McLeod (TAC Cup) will also miss.
Key defender Kym Eagleson (unavailable) and promising teenager Paddy Anderson (under 18s) are among the Roosters' outs.
Camperdown boosted its spine for its battle against arch rival Cobden, bringing in key defender Clinton Fitzgerald and forward Liam Darcy.
Cobden added forward Chris Paatsch, running player Jake Evans and first-gamer Charlie Darcy.
Darcy is the Bombers' 11th senior debutant this season.
Terang Mortlake added creative forward-midfielder Joe Arundell for his first senior game since round one, alongside Damian Moloney, for its must-win game against Koroit.
The Saints will be without Maskell medallist Ben Goodall.
