Mortlake police Acting Sergeant Martin Howells said the car fire was not suspicious and likely started by an electric fault or problem with the engine.
He said a man in his 50s was driving home to Derrinallum about 11pm on Tuesday night when he smelled smoke.
The driver parked on the side of the road before his white 1992 Holden Commodore sedan became fully engulfed in flames.
The Mortlake Country Fire Authority unit attended and put out the fire.
The vehicle valued at about $2500 was completely destroyed.
Traffic in Colac was interrupted at the Princes Highway cutting on the east of the city from 6.45am Wednesday.
Police reported a prime mover and semi-trailer had broken, blocking the west-bound lanes. Police directed traffic while heavy haulage was called in to move the vehicle.
