Police at the scene said the Ryan's B-double truck, carrying two containers, was travelling on Cobden-Warrnambool Road towards Cobden about 3.40am.
They believed the driver may have approached the tight left-bend too fast at the intersection of Maddens Bridge Road, possibly due to the heavy fog.
The vehicle rolled onto the driver's side on the Cobden side of the bend.
The male driver was pronounced dead at the scene and was the only occupant of the truck.
Police from Timboon are investigating the circumstances surrounding the incident, which happened about a kilometre south of the crash that killed a 31-year-old Cobden man on January 15.
Detours were put in place at the Fonterra factory in Cobden, diverting traffic via the Cobden-Terang Road onto the Cobden-South Ecklin Road.
Traffic from Warrnambool was being diverted onto the Timboon-Terang Road onto the Cobden-South Ecklin Road through Cobden.
Police urged motorists to be cautious and drive according to weather conditions.
The fatality brings Victoria's provisional road toll to 36 for 2015, compared to 2014.
