Police investigators said the male motorcyclist, 36, was heading north along Foxhow Road, south-west of the twin lakes, about 12.55am.
Riding a Victory motorcycle, he crossed on to the wrong side of the road, just past the Bakers Road intersection, and collided with a south-bound car. The rider, who was believed to have been heading home, died at the scene.
The sedan was carrying four people, all from Warrnambool.
A woman in her 20s, who was the front seat passenger in the car, suffered a minor laceration to her head and was taken to Camperdown hospital. She was released shortly afterwards.
The other occupants of the car, all men believed to be in their 20s, were not injured.
An Ambulance Victoria spokesman said they were checked by paramedics at the scene but did not require medical assistance.
He said the emergency helicopter was activated when the accident was reported but was sent back to base a short time later.
Senior Constable Marty Howells, of Camperdown police, said the collision occurred on a straight stretch of road and in good weather conditions. He said investigations into the cause of the crash were continuing.
Meanwhile, a luxury car reached speeds up to 220km/h during a short police pursuit near Tyrendarra on Saturday. A highway patrol unit detected the black BMW four-wheel-drive speeding along the Princes Highway about 35 kilometres east of Portland at 1pm.
Police attempted to intercept the vehicle, which was heading east towards Port Fairy, but called off the chase after just one minute.
They were concerned about the potential danger to the heavy traffic using the highway at the time.
The vehicle was detected at 220km/h before the pursuit ended.
Police were unable to obtain number plate details.
Units from neighbouring areas were dispatched in an effort to intercept the BMW but it was not seen again.
Police believe the driver may have turned off the highway on to minor roads in an effort to evade them.
Portland police urged anyone who may have seen the vehicle to contact them or Crime Stoppers on 1800 333 000.
