The 50-year-old was last seen leaving a relative's property in Hamilton in a silver 2007 Holden Rodeo Ute, headed toward Warrnambool just before 7pm on Sunday.
Police said Mr Burn's disapearance "was baffaling".
Detective Sergeant Mark James, of the Hamilton police crime investigation unit, said there had been no sighting of Mr Burn or his vehicle since Sunday night.
He said Mr Burn had not been in touch with family or friends and his bank account had not been touched.
Detective Sergeant James said Victoria Police aircraft had flown over the likely routes Mr Burn would have taken from Tarrington to fill-up with fuel in Hamilton and then from Hamilton back to his Warrnambool home.
"Mr Burn has just never arrived home. We have had police officers drive those routes to see if there are any signs of a vehicle leaving the road or that the vehicle broke down," he said.
"We have no new leads. This is very out of the ordinary for Mr Burns. He spent the weekend helping his brother do home renovations and just didn't turn up at home on Sunday night."
Detective Sergeant James said Mr Burn was hearing impaired but had no other known issues.
Mr Burn is described as 180cm tall with a thin build and short brown hair. He was last seen wearing a hi-vis jacket, blue work pants and work boots.
The registration of the vehicle Mr Burn was driving is WLV-615.
Anyone who sees Mr Burn or has any information concerning his whereabouts is urged to call triple zero (000) immediately.
