The truck driver received minor injuries when the semi-trailer rolled over about 3.45am on Friday on the Princes Highway at Stonyford, west of Colac.
A police spokeswoman said the accident happened when the unloaded truck was travelling east.
The spokeswoman said the driver lost control and swerved to the incorrect side of the road.
The spokeswoman said the truck rolled over a guard rail on the side of the highway and came to rest on the driver's side door.
A heavy haulage vehicle is to be used to retrieve the semi-trailer and VicRoads is managing traffic around the accident site.
