The crash happened on the Princes Highway, approximately 100 metres from the Glenelg River Bridge, just before 5.30am.
Police believe the B-Double truck carrying logs was travelling east along the highway when he collided with a tractor travelling west.
Both vehicles burst into flames, with the truck pushing the tractor back onto the bridge.
The tractor driver died at the scene and is still to be identified.
The driver of the truck, a 55-year-old man, was conveyed to the Mt Gambier with serious injuries.
The highway is expected to be closed all day and motorists are asked to avoid the area.
Westbound traffic is being diverted to the Glenelg Highway at Casterton and Winnap-Nelson Road.
Eastbound traffic is being diverted via the Portland-Nelson Road in Mount Gambier.
